background: RAP2B belongs to a family of RAS-related GTP-binding proteins. RAP proteins share approximately 50% amino acid identity with the classical RAS proteins and have numerous structural features in common. However, at their 61st amino acid the glutamine in RAS proteins is replaced by threonine in RAP proteins. RAP2A interacts with phospholipase C, epsilon 1 (PLCE1). Function: Small GTP-binding protein which cycles between a GDP-bound inactive and a GTP-bound active form. Involved in EGFR and CHRM3 signaling pathways through stimulation of PLCE1. May play a role in cytoskeletal rearrangements and regulate cell spreading through activation of the effector TNIK. May regulate membrane vesiculation in red blood cells. Subunit: Interacts with PLCE1. Interacts with SGSM1, SGSM2 and SGSM3. The GTP-bound form of RAP2B interacts with RUNDC3A Subcellular Location: Recycling endosome membrane; Lipid-anchor; Cytoplasmic side. Note=Associated with red blood cells-released vesicles Tissue Specificity: Expressed in red blood cells Post-translational modifications: Palmitoylated. Palmitoylation is required for association with recycling endosome membranes and activation of TNIK Similarity: Belongs to the small GTPase superfamily. Ras family.
