Plated and cultivated Talquetamab at 4℃ overnight, then with the addition of NF-AT-Jurkat cells for 6 hours. Bright-Lite was used to detect the fluorescent signal. As shown in fig, Talquetamab was able to activate the NF-AT signaling pathway, and the EC50 was 7.499 nM.
Talquetamab bound to huGPRC5D-HEK293 cells, and then rebounded to fluorescent secondary antibodies (Anti-human IgG, Fcγ PE) , and test by flow cytometry. As shown in fig 3, Talquetamabbound to huGPRC5D-HEK293 cells, and the EC50 was 2.067 nM.
Talquetamab bound to huCD3e-jurkat cells, and then rebounded to fluorescent secondary antibodies (Anti-human IgG, Fcγ PE) , and test by flow cytometry. As shown in fig, Talquetamab bound to huCD3e-jurkat cells, and the EC50 was 6.175 nM.
Talquetamab bound to GPRC5D protein, and then rebounded to secondary antibodies (Anti-human-IgG-Fc-HRP) , and read OD450. As shown in fig, Talquetamab bound to hu-GPRC5D-VLP, and the EC50 was 0.978 nM.
The purity of Anti-GPRC5D & CD3 Reference Antibody (Talquetamab) is more than 97.10%, determined by SEC-HPLC.
Anti-GPRC5D & CD3 Reference Antibody (Talquetamab) on SDS-PAGE under reducing (R) condition. The purity of the protein is greater than 95%.
