Super oxide anions generated as the byproduct of Xanthine Oxidase catalyzed Xanthine oxidation and the anion can oxidize hydroxylamine to nitrite which appears to be amaranth purple in the presence of chromogenic agent and thus the absorbance at certain wavelength can be detected by spectrophotometer. The presence of SOD in the system specifically inhibits the oxidization caused by super oxide anions and fewer nitrite anions generated. This would lower the absorbance of the sample tube compared to the reference tube without SOD and the SOD activity can be calculated with the formula given.There are three types of SOD in plant tissues: CuZn-SOD, Mn-SOD and Fe-SOD. The SODs in simple plants are mainly composed of Mn-SODs or Fe-SODs while the SODs in higher plants are normally CuZn-SODs. Also, most of the CuZn-SODs in plant tissues can be found inside the cytoplasm or the chlorophyll and mitochondria contains Mn-SODs while Fe-SODs are existed in the chlorophyll of some species of plants. The total SOD activity is defined as the activity summation of each SOD. To remove both Mn-SOD and Fe-SOD activities in the system, plant should be pre-treated before measurement and CuZn-SOD activity remains unchanged. 植物体中SOD可分为3 种类型:即铜锌-SOD(CuZn-SOD)、锰-SOD(Mn-SOD)和铁-SOD(Fe-SOD)。低等植物以Fe-SOD 和Mn-SOD 为主,高等植物以CuZn-SOD为主,CuZn-SOD主要位于细胞质和叶绿体中,Mn-SOD主要位于线粒体中,Fe-SOD一般位于一些植物的叶绿体中。三者相加等于总SOD(T-SOD)。经样本前处理过的样本中Mn-SOD和Fe-SOD活力丧失,但CuZn-SOD活力不变。 |
