The protein encoded by this gene belongs to the highlyconserved cyclin family, whose members are characterized by adramatic periodicity in protein abundance through the cell cycle.Cyclins function as positive regulators of cyclin-dependent kinases(CDKs), and thereby play an essential role in the control of thecell cycle. Different cyclins exhibit distinct expression anddegradation patterns, which contribute to the temporal coordinationof each mitotic event. Studies of similar genes in chicken anddrosophila suggest that this cyclin may associate with CDC2 andCDK2 kinases, and may be required for proper spindle reorganizationand restoration of the interphase nucleus. Alternatively splicedtranscript variants encoding different isoforms have been describedfor this gene. Function: Cyclins are positive regulatory subunits of thecyclin-dependent kinases (CDKs), and thereby play an essential rolein the control of the cell cycle, notably via their destructionduring cell division. Its tissue specificity suggest that it may berequired during early meiotic prophase I. Subunit: Interacts with CDK2 kinase. Subcellular Location: Nucleus. Tissue Specificity: Testis specific. In testis, it is expressed indeveloping germ cells, but not in Leydig cells. Weakly or notexpressed in other tissues. Post-translational modifications: Ubiquitinated (Probable). Ubiquitination leads to itsdegradation during anaphase entry, after degradation of CCNB1. Similarity: Belongs to the cyclin family. Cyclin AB subfamily. Tissue/cell: rat testis tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer ( 0.01M, pH 6.0 ), Boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer (normal goat serum,C-0005) at 37℃ for 20 min;
Incubation: Anti-Cyclin B3 Polyclonal Antibody, Unconjugated(bs-7884R) 1:200, overnight at 4°C, followed by conjugation to the secondary antibody(SP-0023) and DAB(C-0010) staining
Blank control: Hela. Primary Antibody (green line): Rabbit Anti-Cyclin B3 antibody (bs-7884R) Dilution: 1μg /10^6 cells; Isotype Control Antibody (orange line): Rabbit IgG . Secondary Antibody : Goat anti-rabbit IgG-PE Dilution: 3μg /test. Protocol The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 90% ice-cold methanol for 20 min at-20℃. The cells were then incubated in 5% BSA to block non-specific protein-protein interactions for 30 min at at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.
