产品介绍 DAB (3,3′-Diaminobenzidine; 3,3′,4,4′-Biphenyltetramine; 3,3′,4,4′-Tetraaminobiphenyl) is Peroxidase substrate suitable for use in immunoblotting and immunohistological staining procedures. This substrate produces an insoluble end product that is brown in color and can be observed visually. The kit contains three components: the DAB Chromogen, Substrate Buffer and Staining Assistant. All components are in liquid form. This new 3-Solution DAB product has improved stability, ease of formulation, and greater sensitivity than previous DAB products.

DAB,即3,3’-二氨基联苯胺(3,3’-diaminobenzidine)是辣根过氧化物酶(Peroxidase)的常用生色底物,在过氧化氢的存在下失去电子而呈现出颜色变化,形成浅棕色不溶性产物。用于检测过氧化物酶的活性,它灵敏度高,特异性好,是HRP结合物最常用的底物,常在免疫组化,原位杂交,Western blot等膜显色或检测细胞或组织内源性的过氧化物酶中应用广泛。
用法:
1. 临用前配制 10 mL 1×DAB 显色工作液:将 0.5 mL 20× DAB 显色液 A 加入 9 mL 1×PBS 中并涡旋混匀。向 DAB / PBS 混合物中加入0.5 mL 20×DAB显色液B涡旋混合均匀。 可以根据需要按比例缩放体积。 注意:不要使用含有叠氮化钠的缓冲液,叠氮化钠是一种HRP抑制剂。
2. 在样品的最后一次洗涤完毕后,去除洗涤液,加入足够 的 DAB 显色工作液,确保能充分覆盖样品。室温避光孵育 3-30 分钟或更长时间,直至显色达到预期深浅。免疫印迹实验,应保证样品在一个足够多的染色液中且能被自由晃动。
3. 去除 DAB 染色工作液,用 PBS 冲洗样品几次以中止显色反应。对于用显微镜观察的细胞或组织样品,可复染样品(可选)并安装在水性封固介质中。或者,可以将样品脱水并安装在有机封固剂中。免疫印迹实验,可将样品用水冲洗,风干,然后在室温下储存。

储存与运输:
-20℃避光密闭保存,一年有效。避免反复冻融。
2~8℃运输,在途时间5天对试剂无影响。

注意事项
1. DAB对人体有害,操作时请小心,并注意有效防护以避免直接接触人体或吸入体内。
2. 显色工作液应现用现配,新鲜配制的工作液应为无色或浅棕色,如颜色过深,请勿使用。  
3. 显色时间严格控制,根据情况调整,以免显色过度。固相膜显色数小时后即会褪色,不能永久存在。
4. 请将沾有DAB显色液A的容器等放在含有3% KMnO4,2% NaHCO3 的溶液浸泡3h,以减少污染。 
5. 本产品仅限于专业人员的科学研究用,不得用于临床诊断或治疗,不得用于食品或药品,不得存放于普通住宅内。
