FAF1 (Fas-associated protein factor 1) specifically interacts with the cytoplasmic domain of wild type, but not mutated, Fas in both yeast and mammalian cells. It is transiently expressed in T cells and potentiates Fas-induced apoptosis. FAF1 lacks a DDH (Death domain homologous region), in contrast to TRADD, FADD, and RIP, and cannot induce apoptosis independent of Fas activation. Fas associated factor 1; CGI ; FAF1 protein; Fas (TNFRSF6) associated factor 1; FAS associated factor 1; FLJ37524; hFAF1; HFAF1s; Protein FAF1; TNFRSF6 associated factor 1. Function: Potentiates but cannot initiate FAS-induced apoptosis. Subunit: Specifically interacts with the cytoplasmic domain of FAS. Interacts with NLRP12 DAPIN/PYRIN domain via its UBA domain. Subcellular Location: Nucleus. Tissue Specificity: Most abundant in testis, slightly less abundant in skeletal muscle and heart, followed by prostate, thymus, ovary, small intestine, and colon. Not detected in the peripheral blood leukocytes. Similarity: Contains 1 UBA domain. Contains 1 UBX domain. Paraformaldehyde-fixed, paraffin embedded (rat colon); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (FAF1) Polyclonal Antibody, Unconjugated (bs-1348R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
Tissue/cell: Rat testis tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer ( 0.01M, pH 6.0 ), Boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer (normal goat serum,C-0005) at 37℃ for 20 min;
Incubation: Anti-FAF1 Polyclonal Antibody, Unconjugated(bs-1348R) 1:200, overnight at 4°C, followed by conjugation to the secondary antibody(SP-0023) and DAB(C-0010) staining
