一、服务内容:
本服务项目主要为接收客户的抗体、蛋白、多肽、小分子化合物进行常规标记物标记。

2. 客户提供待标记样品,我公司按照合同要求安排标记服务,并在规定时间内完成标记委托。
3. 标记完成后,我公司按照客户要求分装,免费低温邮寄给客户,同时提供标记报告。
三、样品要求:
1. 待标记样品需保证90%以上的纯度,样品中不含有干扰标记物结合的成分,如有特殊成分,请提前标注说明。
2. 待标记样品最小标记量为1mg,最好是干粉,如果是液体,浓度应大于1mg/ml。
3. 抗体IgG样品中应不含BSA、叠氮钠、甘氨酸等成分。
4. 大分子蛋白应提供分子量、等电点、缓冲液成分及pH等信息;小分子多肽需提供氨基酸序列;小分子化合物还需提供分子结构式。
5. 请提前说明任何特殊情况。

五、标记物介绍:
碱性磷酸酶(alkaline phosphatase,ALP或AP)是免疫诊断试剂产品最常用的标记酶之一,优点是稳定性高、灵敏度高。ALP是一种能够将对应底物去磷酸化的酶,即通过水解磷酸单酯将底物分子上的磷酸基团除去,并生成磷酸根离子和自由的羟基,这类底物包括核酸、蛋白、生物碱等。而该脱去磷酸基团的过程被称为去磷酸化或脱磷酸化。碱性磷酸酶是磷酸酶的一种,磷酸酶的作用与激酶的作用正相反,激酶是磷酸化酶,可以利用能量分子,如ATP,将磷酸基团加到对应底物分子上。
辣根过氧化物酶(Horseradish Peroxidase,HRP)活性高,稳定,分子量小,纯酶容易制备,所以最常用。HRP广泛分布于植物界,辣根中含量高,它是由无色的酶蛋白和棕色的铁卟啉结合而成的糖蛋白,糖含量18%。HRP由多个同功酶组成,分子量为40,000,等电点为PH3~9,酶催化的最适PH因供氢体不同而稍有差异,但多在PH5左右。酶溶于水和58%以下饱和度硫酸铵溶液。HRP的辅基和酶蛋白最大吸收光谱分别为403nm和275nm,一般以OD403nm /OD275nm的比值RZ(德文ReinheitZahl)表示酶的纯度。高纯度的酶RZ值应在3.0左右(最高可达3.4)。RZ值越小,非酶蛋白就越多。
FITC和RhodamineB(RBITC)是常见的荧光素,性质较稳定;
Cy系列荧光染料由于荧光较强,对光猝灭有抵抗力而荧光不宜消退,近年来应用比较广泛;
