Background:
Epithelial-mesenchymal transition (EMT) is a biological process that allows epithelial cell to assume mesenchymal phenotype. EMT and its reverse process, mesenchymal-epithelial transition (MET), are originally defined in the context of developmental stages and re-engaged in adults during wound healing, tissue regeneration, organ fibrosis, and cancer progression. Epithelial cells normally interacts with basement membrane via its basal surface. During EMT, they lose their cell polarity and cell-cell adhesion, and gain mesenchymal cell phenotype, including enhanced migratory capacity, invasiveness, elevated resistance to apoptosis, stem-like features, and increased production of ECM components. Accordingly, there is reprogrammed gene expression with EMT. Especially, some cytoskeleton proteins (such as cadherin family members), tight-junction proteins (such as Occludin and ZO-1) and intermediate filament of mesenchymal origin (such as Vimentin) undergo significant changes and serve as best EMT markers to evaluate EMT.
